چکیده

A LDOHEXOSES REACT WITH o-toluidine in the presence of glacial acetic acid to produce a green color, which follows Beer’s law over a wide range of concentrations. This reaction is relatively selective for glucose. An improvement in manual procedures involving this reaction (1, 2) was reported by Feteris (3), who determined glucose directly on serum without deproteiization. She also improved the sensitivity of the o-toluidine reagent by adding thiourea. An automated o-toluidine method for glucose requiring dialysis was reported by Zender (4). This paper reports an automated procedure for determining glucose in serum or plasma, without dialysis, using an o-toluidine reagent. The new automated procedure is compared with the manual method of Feteris (3).

Method for simultaneous direct estimation of glucose and xylose in serum.

A direct procedure for measuring serum monosaccharides is presented. Borate intensifies the colors produced when these substances react with o-toluidine, which, in the case of glucose and xylose, have absorption maxima at 480 nm and 630 nm, respectively. Thus, these sugars may be determined singly or simultaneously in a single sample by differential spectrophotometry. An alternative technique i...

An o-toluidine method for body-fluid glucose determination.

Featured Article: Dubowski KM. An o-toluidine method for body fluid glucose determination. Clin Chem 1962; 8:215–35. This report described a simple, rapid, and versatile method for manual measurement of glucose in whole blood, serum, cerebrospinal fluid (CSF), and other body fluids, with results fully compatible with the then–widely used automated Hoffman procedure for body-fluid glucose: reduc...

Evaluation of a glucose oxidase-peroxidase method adapted to the single-channel autoanalyzer and SMA 12-60.

The glucose oxidase-peroxidase method for determining serum glucose, with the ammonium salt of 2,2’-azine-di (3 ethyl benzothiazoline-(6)-sulfonic acid) as the chromogen, was adapted to use with the single-channel AutoAnalyzer and the SMA 12/60 (Technicon Corp.). The extended linearity (up to 500 mg/100 ml on both systems) eliminated the need for numerous dilutions in the 300 to 500 mg/100 ml r...

Rate of Glucose Utilization by Blood Cells in Serum and Plasma Specimens With or Without Using Preservative

Objective: This study aims to estimate and compare the time-course change in blood glucose levels by blood cells in serum, and plasma with or without preservatives, which may reflect the rate of glucose utilization by blood cells. Method: This laboratory-based cross-sectional study was carried out using a blood specimen of 28 participants among which 14 were diagnosed with diabetes and 14 were...

Direct Automated Determination of Glucose by a Glucose Oxidase-peroxidase System.

THE DETERMINATION of glucose by the glucose oxidase-peroxidase system has been simplified by Hill and Kessler (2) with the use of an automated system first described by Skeggs (1). Wincy and Marks (3) have also developed a method using o-tolidine in the automated system. These procedures require a large quantity of sample and use of a dialyzer to separate the protein before color development. T...
